Changes in Enzyme Activities, Nucleic acid and Protein Centents Of Brains in Neuroencephalopathy induced experimentally by Neurotoxic Compounds



Abstract
Neuroencephalopathy could be induced experimentally by various neurotoxic agents. In order to investigate biochemical changes observed in functional disturbances in brains, activities of enzymes involved in glutamte and electrolyte metabolisms, concentrations of nucleic acids and proteins, and water contents were determined in brains of rats with neuroencephalopathy induced by the administration of ammonium chloride, penicillin, ouabain or gamma-aminobutyrate(GABA), and were compared with those of normal rats.
1)Significant increases in blood and brain amminia and decrease in brain glutaminase activity were observed to be restricted to the rats with ammonia toxicity induced by the administration of ammonium chloride.
2)Significant changes in activities of brain glutamine synthetase and glutamate dehydrogenase were found in the rats with neuroencephalopathy induced by the administration of ammonium chloride or penicillin.
3)A significant dectease in the activity of brain Na??,K?? -ATPase was observed in the rats with neuroencephalopathy induced by the administration of ammonium chloride, penicillin or ouabain. Significant changes in Na??, K?? and water contents of brains were, however, observed to be limited to the rats with neuroencephalopathy induced by the administration of penicillin or ouabain,and the degree of changes in the enzyme activity and the electrolye contents was shown to be greatest in the rats with ouabain-induced encephalopathy among the rats with the experimentally induced neuropathy studied.
4)While no significant changes in DNA and RNA contents of brains were observed in the rats with the experimentally induced encephalopathy studied in the present work, protein content was found to be decreased in brains of rats with penicillin or ourbain-induced neuroencephalopathy.
5)In the rats with neuroencephalopathy inducde by the administration of GABA, activities of enezymes involved in glutamate and electrolyte metabolisms, concentrations of electrolytes, nucleic acids and proteins, and water contents in brains were unchanged.
The results obtained by the present study revealed that ammonium chlorideinduced encephalopathy might result primarily from disturbance of ammoniaflutamate-glutamine metabolism, penicillin encephalopathy from disturbances of both glutamate-glutamine and electrolyte metabolisms and that ouabain induced epileptiform encephalopathy parimarily from disturbance of electrolyte metabolism.It can not be ruled out, however, that neuroencephalopathy studied in the present work might be associated with disturbance of energy metabolism in the brains.
